PDPK1
MIM.605213 16p13.3
Isoforms of AKT (protein kinase B or PKB or AKT1) (MIM.164730) are overexpressed in some ovarian, pancreatic, and breast cancer cells, and PKB has been shown to protect cells from apoptosis.
Activation of AKT1 is preventable by inhibitors of phosphoinositide 3-kinase (PI3K). AKT1 is stimulated by insulin or growth factors after phosphorylation of PKB at thr308 and ser473.
PDK1 phosphorylates PKB at thr308 in response to phosphatidylinositol 3,4,5-trisphosphate (PtdIns(3,4,5)P3) or phosphatidylinositol 3,4-bisphosphate (PtdIns(3,4)P2) and enhances AKT1 (PKB) activity.
556-amino acid PDPK1 protein contains a catalytic domain with 11 classic kinase subdomains and a C-terminal pleckstrin homology (PH) domain.
