Kruppel-like factor 6 (KLF6) is a ubiquitous zinc finger tumor suppressor gene that is often mutated in prostate cancer.
KLF6 encodes a transcription factor that has many target genes, including TGF-β and TGF-β receptors. KLF6 is mutated in more than 70% of primary prostate cancers.
KLF6 inhibits cell proliferation by increasing the transcription of the Cip/Kip cell-cycle inhibitor p21, independent of p53. Mutation of the gene in tumor cells eliminates the cell-cycle-blocking activity of p21.
