DLLs are human homologs of the Drosophila Notch ligand Delta. Notch signaling is essential for appropriate cell differentiation and cell fate decisions. Two Notch pathway elements, NOTCH3 (MIM.600276) and JAG1 (MIM.601920), are implicated in the Alagille (MIM.118450) and CADASIL (MIM.125310) congenital syndromes, respectively, and constitutively active aberrant forms of Notch are associated with human cancers.
