Biobanks of fresh, unfixed human tissue represent a valuable source for gene expression analysis in translational research and molecular pathology.
Minimal RNA degradation was detected after 30 minutes of thawing in unfixed samples. This degradation was accompanied by relevant changes in gene expression for FOS and BCL2 at 45 minutes.
Modified primer design or the use of different housekeeping genes could not rectify the changes for FOS.
Repetitive thawing cycles had similar effects on (...)

Neoplasms develop through acquisition of capabilities that involve tumour cell aspects and modified microenvironment interactions, resulting in unrestricted growth due to a stepwise accumulation of cooperative genetic alterations that affect key molecular pathways.
The correlation of these molecular aspects with morphological changes is essential for better understanding of essential concepts as early neoplasms/precancerous lesions, (...) -
fluorescence polarization assay
25 November 2011Currently, the most used methods for genotyping of SNPs are costly and time consuming. It is necessary to develop a method that is more accurate, cost-effective, and simple.
An improved fluorescence polarization assay based on asymmetric polymerase chain reaction hybridization for screening SNPs has been developed.
Studies show the accuracy, simplicity, and cost-effectiveness of the fluorescence polarization assay in the detection of SNPs.
In a study, the fluorescence polarization assay (...) -

microfluidic device
9 March 2010The dynamics governing gene regulation have an important role in determining the phenotype of a cell or organism.
From processing extracellular signals to generating internal rhythms, gene networks are central to many time-dependent cellular processes.
Recent technological advances now make it possible to track the dynamics of gene networks in single cells under various environmental conditions using microfluidic ’lab-on-a-chip’ devices, and researchers are using these new techniques to (...) -
RNAi screening and cancer
21 May 2009RNAi screening and cancer
Various types of cancers are generated through mutations or dysregulations of oncogenes/tumor suppressor genes involved in cell cycles and signaling transduction pathways.
To identify cancer therapeutic targets whose inhibition selectively kills cancer cells, synthetic lethal screening is being developed to identify genes whose intervention suppresses tumor progression only when combined with the dysregulation of the genes.
The recent emergence of genomic (...) -
in silico gene discovery
14 July 2008Complex diseases can involve the interaction of multiple genes and environmental factors. Discovering these genes is difficult, and in silico based strategies can significantly improve their detection.
Data mining and automated tracking of new knowledge facilitate locus mapping. At the gene search stage, in silico prioritization of candidate genes plays an indispensable role in dealing with linked or associated loci.
In silico analysis can also differentiate subtle consequences of coding (...)
