classic cloning
The functional cloning, or classic cloning, approach has been successfully used to study a variety of inborn errors of metabolism, such as phenylketonuria and disorders of hemoglobin synthesis. Common to these genetic diseases is knowledge of the abnormal gene product and the corresponding protein. When the affected protein is known, a variety of methods can be employed to isolate the normal gene, to clone it, and ultimately to determine the molecular changes that affect the gene in patients with the disorder.
